Abstract (en)
The subjects of this study was the phytochemical content of autochthonous and nostrificated apple
cultivars (Malus domestica Borkh.) from the territory of Serbia. For comparison, commercial
international standard cultivars and resistant apple cultivars were also analysed. All samples were
obtained from the same locality, fruits (separated into mesocarp and peel) and leaves were analysed.
The evaluated phytochemical parameters (sugar, organic acid, element, and phenolic content) were
assessed with the aim to express the positive properties of autochthonous apple cultivars and to
create a foundation for further studies. Also, some of the used analytical methods were optimised.
Quantification of individual compounds was performed with HPAEC-PAD, HPAEC-CD, ICPOES,
ICP-QMS and UHPLC-DAD MS/MS methods. Phenolic profile in mesocarp of selected
cultivars was obtained by the usage of high-performance liquid chromatography coupled with mass
spectrometry, with LTQ OrbiTrap XL MS system. Antimicrobial activity in mesocarp, peel, and
leaves was assessed with the employment of bioautography. The separation of selected extracts was
repeated to fractionate the extracts correspondingly to the delineated inhibition zones on the TLC
plate. Detailed characterisation of the compounds present in the fractions was performed with the
usage of UHPLC QToF MS system.
Obtained results of sugars and organic acid quantification in mesocarp are implying that some
autochthonous cultivars have potential to be used in the same manner as commercial dessert apple
cultivars. The analysis of elemental content does not indicate their influence on separation of
samples by variety, although there are differences dependent on the tissue type. Phloretin, phlorizin,
5-O-caffeoylquinic acid, kaempferol, and p-coumaric acid are present in significantly higher levels
in the autochthonous cultivars, in comparison to the examined standard and resistant cultivars.
Higher total phenolic content and antioxidant activity was also determined in autochthonous
cultivars. Obtained results of Peel and leaf extracts have shown antimicrobial activity against the
investigated bacteria strains (Escherichia coli, Staphylococcus aureus, Bacillus subtillis and
Erwinia amylovora). In the growth-inhibition zones presence of triterpenoids, coumaoyl-, and
coffeoylquinnic acids, their glycosides, and phloretin glycosides and acylated derivatives was
determined. Some acylated phloretin derivatives were described for the first time in apple leaves.

Predmet ove disertacije je fitohemijski sastav autohtonih i odomaćenih sorti jabuke (Malus
domestica Borkh.) sa teritorije Republike Srbije. Radi poređenja ispitane su i alohtone sorte –
komercijalne internacionalne standardne sorte i rezistentne sorte. Ispitivani uzorci su prikupljeni sa
istog lokaliteta. Ispitivani su plod (razdvojen na mezokarp i pokožicu) i list. Analizirani fitohemijski
parametri (sadržaj šećera, organskih kiselina i elemenata, kao i ispitivanje sadržaja i profila fenolnih
jedinjenja) su razmotreni u cilju isticanja pozitivnih osobina autohtonih sorti i postavljanja temelja
za dalje studije. Takođe su optimizovane neke od upotrebljenih analitičkih metoda.
Pojedinačna jedinjenja su kvantifikovana primenom HPAEC-PAD, HPAEC-CD, ICP-OES, ICPQMS
i UHPLC-DAD MS/MS metoda. Fenolni profil je ispitivan kod mezokarpa odabranih
uzoraka, primenom tečne hromatografije spregnute masenom spektrometrijom, LTQ OrbiTrap XL
MS sistemom. Primenom bioautografije ispitana je antimikrobna aktivnost ekstrakata mezokarpa,
pokožice i lista jabuke. Zatim je razdvajanje odabranih ekstrakata ponovljeno da bi ekstrakti bili
frakcionisani ekstrakcijom iz zona definisanih prema antimikrobnoj aktivnosti na razvijenoj TLC
ploči. Detaljna karakterizacija jedinjenja u dobijenim frakcijama je obavljena primenom UHPLC
QToF MS tehnike.
Rezultati kvantifikacije sadržaja saharida i organskih kiselina u mezokarpu ukazuju da pojedine
autohtone sorte mogu da nađu sličnu primenu kao i standardne desertne sorte. Analiza sadržaja
elemenata ne ukazuje njihov uticaj u razdvajanju uzoraka po sortama, ali postoje razlike prema tipu
tkiva. U poređenju sa ispitivanim standardnim i rezistentnim sortama, kod autohtonih sorti u većoj
količini su prisutni floretin, florizin, 5-O-kofeoilkininska kiselina, kamferol, i p-kumarinska
kiselina. Primetan je i veći sadržaj ukupnih fenola i veća antioksidativna aktivnost kod autohtonih
sorti. Metanolni ekstrakti pokožice i lista su pokazali antimikrobnu aktivnosti na ispitivanim
sojevima bakterija (Escherichia coli, Staphylococcus aureus, Bacillus subtillis i Erwinia
amylovora), pri čemu je u inhibicionim zonama ustanovljeno prisustvo triterpenoida, kumaroil- i
kefeoilkininskih kiselina i njihovih metil- derivata, kvercetina i njegovih glikozida, floretina i
njegovih glikozida, kao i acilovanih derivata floretina, koji su prvi put opisani u listu i plodu jabuke.
